![]() d) Represents Western blot detection of β-dystroglycan (β-DG) in C57BL/10 and mdx nerve extracts. c) C-ter utrophin antibody (K7) detection showed the full utrophin (Up395) in C57BL/10 and both Up395 and the short product Up71 (71 kDa) in mdx extract. b) Dp116 detection on the same extracts with C-terminal dystrophin antibody (H4). a) Western blots revealed a dystrophin isoform (Dp116) and utrophin isoforms (Up395 and Up71) in C57BL/10 and mdx total nerve extracts, using a monoclonal antibody recognizing both dystrophin and utrophin (5A3). ( A) Immunodetection of DAP complex in C57BL/10 and mdx peripheral nerve extracts. We proposed that these two proteins could be implicated in Schwann cell proliferation in response to a microenvironment stress such as mediated by accumulating macrophages in mdx mouse muscle inflammation sites. Our results highlight the potential participation of the short utrophin isoform and the cleaved form of beta-dystroglycan in mdx Schwann cell membrane architecture. In addition, this utrophin isoform (Up71) seems to have greater affinity to the 30 kDa beta-dystroglycan which could explain the increased stabilization of this 30 kDa form at the membrane compartment. Moreover, we showed for the first time that the short isoform of utrophin (Up71) was over-expressed in mdx Schwann cells compared with wild-type. Our results showed that Dp116 had greater affinity to the full length form of beta-dystroglycan than the 30 kDa form. This activation leads to an accumulation of the 30 kDa beta-dystroglycan isoform and has an impact on the anchorage of Dp116 and utrophin isoforms in mdx Schwann cells membrane. We showed that metalloproteinase-9 was more activated in mdx nerve than in wild-type ones. Here we asked if the processing of the beta-dystroglycan could influence the anchorage of Dp116 and/or utrophin in normal and mdx Schwann cell membrane. Metalloproteinase creates a 30 kDa fragment of beta-dystroglycan, leading to a disruption of the link between the extracellular matrix and the cell membrane. In peripheral nerve, matrix metalloproteinase activity disrupts the dystroglycan complex by cleaving the extracellular domain of beta-dystroglycan. sarcoglycans, and alpha- and beta-dystroglycan, which anchor the cytoplasmic protein subcomplex to the extracellular basal lamina. Dp116 is associated with multiple glycoproteins, i.e. NOTE: All combinations may not be valid, check spec sheet for valid part numbers. In the peripheral nervous system, utrophin and the short dystrophin isoform (Dp116) are co-localized at the outermost layer of the myelin sheath of nerve fibers together with the dystroglycan complex. So overall, a really enjoyable listen so far. I take this to be a very positive attribute. Small changes that usually go fairly unnoticed (to me) are more readily apparent. The amp responds very well to EQ though. ![]() The typical hi-fi predicament where much of this sort of source material isn't mixed with a neutral, ultra-resolving audio chain in mind (I feel the same way about the RAALs, TBH). Modern, compressed material is a mixed bag so far.So like, real "euphonics" based on an effective (accurate) rendering of underlying musical intent, as opposed to "euphonic" in the sense of 2nd-order distortion introduced by the signal chain. Also, I get this satisfying sense of euphony as different instruments layer and harmonize. Really shines with acoustic jazz (probably my #1 priority in terms of musical genres) for all the reasons stated above.Like for instance with dense and complex electronic music, where much of the appeal is in texture and 'soundscape'. When listening analytically, there is much to be explored, looking into the music.Separation between instruments is easy to process cognitively. Due to its resolution, doesn't require the volume to be turned up to fully "get" the music (the same kind of thing people say about the Utopia).Lower registers of bass guitar and stand-up bass are well articulated, and their fundamentals are very follow-able. Very well defined without being overdone or hot, just sounds right. The way it renders cymbals is my favorite thing about the amp so far. Imaging is more of the pin-point variety, not the big-expansive-blobs variety. Height - which I usually don't pay much attention to - can be very pronounced with some percussion on occasion, so that's interesting. Depth is decent, though not a stand-out feature probably better than the Bryston SST2's I've heard. I'm hyper-aware of instrument locations on the far ends of the x-axis. ![]() And then I notice, and I think to myself, "Wait, what? I thought this amp was supposed to sound all 'sterile' and sh- so forth." ![]() I catch myself bobbing my head without noticing. This lends it a surprising sense of rhythm. The Susvara connected to the AHB2 reminds me a bit of the RAAL SR1a, actually. It lends the Susvara a different character than what I'm used to, which by itself makes for an interesting listen.
